The Kid, played by Gene Wilder tells Bart when he first meets him that he shoots left-handed, however when he shoots the dynamite to make it explode he uses his right hand.
If you remember correctly, in that same scene when he says he shoots with his left hand, it's obvious that his right hand is steadier. You'll notice that he wears guns on both hips (and uses both of them when the bad guys try to gun down the Sherriff at the railroad construction site), so it's obvious that he is ambidextrous. Maybe he chose to shoot right handed for that particular shot because he needed the extra steadiness of his right hand . . .
