Richard O'Brien has said on a few occasions during interviews that this was a "hint" that Riff isn't what he seems to be.

The hump isn't real. Riff doesn't even have a hump during the part where they undress Brad and Janet because he takes it out of his jacket to wipe off Janet.

Of course it moves! It isn't real!! This can be seen by his excellent posture at the end when he is in his "space" suit!

Mmkay... one might notice that Riff's hump is actually a piece of cotton...

In the movie, part of Riff Raff's character is having a fake hump, that's why it moves around so much.
